Fas/APO-1 promoter polymorphism is not associated with non-melanoma skin cancer.

چکیده
Introduction NMSC incidence is increasing at an alarming rate (1), and the majority of NMSC is likely attributable to UV exposure. An important biological effector of UV exposure is the Fas receptor, which in cooperation with Fas ligand induces apoptosis in keratinocytes after UV exposure (2). The gene encoding Fas, TNFRSF6, contains a promoter single nucleotide polymorphism at position 2670 that disrupts a GAS transcription factor binding sequence (3). Fas mRNA and protein are both induced after UV exposure (4), indicating that the regulation of this gene is important in the cellular response to UV. We tested the hypothesis that the 2670 G/A promoter polymorphism of TNFRSF6 was associated with NMSC.

Background and purpose: Apoptosis is a physiological mechanism of programmed cell death that dysregulation in this process may lead to carcinogenesis. Functional gene polymorphism in Fas can alter transcriptional activities and thus change the risks of cancer. A to G substitution in Fas -670 promoter region disrupts the binding site of STAT1 transcription factor. This study aimed to assess the ...
متن کاملAre there Relationships between the VDR-FokI Polymorphism and Vitamin D and the Insulin Resistance in Non-melanoma Skin Cancer (NMSC) Patients? A Protocol for Case-control Studies
Background and Objectives: Evidence indicate relationships between the sunlight exposure, vitamin D status and non-melanoma skin cancer (NMSC) risk. Vitamin D receptor (VDR) polymorphisms have been linked to metabolic changes in insulin resistance and various cancers. The purpose of this study is to investigate relationships between the sunlight exposure and the NMSC risk. Furthermore, relation...
متن کاملAn examination of the Apo-1/Fas promoter Mva I polymorphism in Japanese patients with multiple sclerosis
BACKGROUND The Apo-1/Fas (CD95) molecule is an apoptosis-signaling cell surface receptor belonging to the tumor necrosis factor (TNF) receptor family. Both Fas and Fas ligand (FasL) are expressed in activated mature T cells, and prolonged cell activation induces susceptibility to Fas-mediated apoptosis. The Apo-1/Fas gene is located in a chromosomal region that shows linkage in multiple scleros...
متن کاملAPO-1/Fas gene: Structural and functional characteristics in systemic lupus erythematosus and other autoimmune diseases
Systemic lupus erythematosus (SLE) is an autoimmune disorder affecting multiple organ systems. It is characterized by the presence of autoantibodies reactive against various self-antigens. Susceptibility to SLE is found to be associated with many major histocompatibility complex (MHC) and non-MHC genes, one of which is APO-1/Fas gene, which is present on chromosome 10 in humans. The APO-1/Fas p...
متن کاملFunctional FAS promoter polymorphisms are associated with increased risk of acute myeloid leukemia.
The FAS (TNFRSF6/CD95/APO-1) gene is silenced in many tumor types, resulting in an inability to respond to proapoptotic signals. The FAS promoter is polymorphic, including a G to A substitution at -1377 bp and an A to G substitution at -670 bp, which occur within SP1 and signal transducers and activators of transcription 1 transcription factor binding sites, respectively. In a case-control stud...
